Key component in mother's egg critical for survivial of newly formed embryo

ScienceDaily (Mar. 29, 2012) ? An international team led by scientists at A*STAR's Institute of Medical Biology (IMB) discovered that a protein, called TRIM28, normally present in the mother's egg, is essential right after fertilisation[1], to preserve certain chemical modifications or 'epigenetic marks' on a specific set of genes. This newly published study paves the way for more research to explore the role that epigenetics might play in infertility.

Previous studies have shown that both nuclear reprogramming as well as 'imprinting' are vital for the survival and later development of the embryo. However, the underlying mechanisms governing the intricate interplay of these two processes during the early embryonic phase have not been clear, until now.

Nuclear Reprogramming

Immediately after fertilisation, the majority of the 'epigenetic marks' on the DNA from the sperm and egg cells are erased. The erasure process, termed nuclear reprogramming, allows the genes from the parents to be reset so that the early embryonic cells can develop into any cell types of the body.

Genomic Imprinting

On the other hand, certain 'epigenetic marks' on a particular set of genes, some from the mother and some from the father must be preserved. These genes are said to be "imprinted" by their parent of origin and preservation of these marks is critical for the survival of the newly formed embryo. Expression of these imprinted genes at the appropriate levels ensures proper development of the embryo. If the 'epigenetic marks' on the 'imprinted' genes are not protected, severe and multiple developmental defects occur in the embryo.

Using genetically identical mice from an inbred mouse strain[2], Drs. Davor Solter and Barbara Knowles, Senior Principal Investigators at IMB, observed that none of the embryos resulting from the fertilisation of eggs lacking TRIM28 survived. The embryos died at varying stages of development, and had very different developmental defects. Given that most genetic diseases attributed to the lack of a single gene have consistently similar defects in every affected individual, the highly varied abnormalities observed in the maternal TRIM28-deficient, yet genetically identical mice, were peculiar.

To further elucidate these findings and the role of TRIM28 during nuclear reprogramming, Dr. Daniel Messerschmidt, the first author of this paper, collaborated with Dr. Anne Ferguson-Smith, a visiting Professor at A*STAR's Singapore Institute for Clinical Sciences (SICS), from the University of Cambridge, UK. Together, they conducted a comprehensive study on the epigenetic state and the level of expression of a group of imprinted genes known to be important for development. Using highly sophisticated microarray analysis and advanced biochemical techniques, they found that not only is the presence of TRIM28 protein important during this critical period, but the timing and amount of TRIM28 made available to the newly formed embryo greatly impacts the expression of these imprinted genes at later developmental stages.

Genomic imprinting was first described by Dr. Davor Solter at the Wistar Insitute and by Dr. Azim Surani at Cambridge University three decades ago. Recently, Dr Solter suggested that the interplay between nuclear reprogramming and genetic imprints could be explained by a simple analogy from school where the teacher writes a long string of formulas on the black board. At the end of the class he marks stretches to be preserved for the next day's lecture (these are the imprinted marks) and asks the cleaning lady to erase the rest (she performs the reprogramming).

Dr Solter said, "For the longest time, we didn't know why some bits of the formulas escaped erasure. Through this study, the mystery is finally revealed! We now know TRIM28 instructs the 'cleaning lady' which part of the formulas she should preserve."

Dr. Azim Surani, who was not involved in this study, said "This elegant study on TRIM28 provides significant mechanistic insights on how maternal factors in eggs ensure faithful epigenetic inheritance that plays a critical role during development. These findings are relevant not only for the widely practised in vitro fertilisation of eggs for the treatment of human infertility, but also more generally for highlighting the importance of epigenetic mechanisms in development and disease."

Dr. Barbara Knowles, the senior author of this paper said, "Lack of TRIM28 in their eggs could explain why some women consistently suffer from multiple failed pregnancies where embryos die at different time points, manifesting multiple, different abnormalities."

Prof Birgitte Lane, Executive Director of IMB, said, "Understanding the role of differential epigenetic modifications is important for the study of human diseases and development. This study has unravelled a critical component essential for nuclear reprogramming. The mechanistic insights revealed have far-reaching clinical implications for treatment of diseases with patient-specific cell therapies. This finding demonstrates how discoveries of fundamental research can shed light on biological questions to open new avenues of formulating therapeutic strategies for the benefits of patients."

Casino Gambling Vote Imminent - Salem, NH Patch

The time has finally come for the N.H. House of Representatives to lay their cards on the table on the issue of casino gambling in the state.

The House will go into session this morning and by the end of business Thursday they must pass House Bill 593 for it to be considered by the state Senate.?

The bill has major implications for Salem as its passage would pave the way for casino gambling at Rockingham Park.

However, Gov. John Lynch has promised a veto of any expanded gambling legislation and it's unclear if the House will get the necessary two-thirds majority to override his veto.

The current bill, which includes licenses for four casinos of varying sizes, was passed out of the Ways and Means Committee by a 14-7 vote.

Rep. Gary Azarian (R-Salem) recently wrote for the majority that residents spend around $80 million annually out of state on gambling and that number could grow to $120 million without action.

"Expanded gaming will keep those dollars here and expand our economy by attracting residents from our neighboring states," Azarian wrote.

The minority opinion was provided by Rep. David W. Hess (R-Hooksett), who questioned if casinos are really what the residents of the Granite State want.

"This bill will almost certainly put a casino within 50 miles of every man, woman and child in?New Hampshire," Hess wrote. "Is that what you want for our state?"

Earlier this week, bill proponents touted the economic impacts, saying it will reduce business taxes while creating jobs.?

?This is the most significant jobs and tax cut bill our Legislature has seen in years,? said?House Ways & Means Chairman Stephen Stepanek.

?Casinos are economic and social black holes," said Granite State Coalition Against Expanded Gambling President Jim Rubens in response. "You don?t solve the problem by making it four times worse by allowing four casinos within easy drive time of almost every town in our state."

Yesterday the N.H. Police Association announced their support of the bill.

Bernanke says US banks could withstand euro shocks

Federal Reserve Chairman Ben Bernanke told a congressional panel Wednesday that U.S. banks could withstand shocks from Europe, even if the debt crisis there significantly worsened.

The U.S. banking system remains exposed to Europe, but stress tests conducted by the Fed found most banks could weather a severe recession triggered by the debt crisis, Bernanke told the House Oversight and Government Reform Committee.

"Although a blow up in Europe would be very costly to the American economy, I think we are in much better shape to meet those challenges than we were a few years ago," Bernanke told the committee.

The Fed looked at how the 19 largest U.S. bank holding companies would handle a recession that drove the unemployment rate to 13 percent and cut stock prices 50 percent. The stress tests found all but four were strong enough to survive.

Bernanke said U.S. financial firms and money market funds have had time to adjust their exposures and hedge their risks. While those exposures have been reduced, Bernanke said that roughly 35 percent of assets in U.S. prime money market funds are European holdings.

The Fed chairman also noted developments that have minimized the danger overseas. Bernanke pointed to bailout support that European leaders provided in exchange for deep budget cuts by the Greek government and he highlighted the agreement by private creditors to reduce Greece's debt.

Treasury Secretary Timothy Geithner told the committee that the Obama administration will not ask Congress for more money to help debt-laden European countries.

Still, Greece's international creditors see "significant risks" that the country might fail to bring down its debt burden within targets, according to a document seen by The Associated Press on Tuesday.

The creditors say Greece's program of austerity measures and structural reforms "could be accident prone," according to the report from the International Monetary Fund, the European Commission and the European Commission.

"Authorities may not be able to implement reforms at the pace envisioned," the report said.

(AP) ? Hines Ward believes he can still play football. The longtime Pittsburgh wide receiver known for his high-wattage smile and his bone-crunching blocks just couldn't stomach the thought of doing it in some strange uniform on some strange field with nary a Terrible Towel in sight.

"I just wouldn't feel right," Ward said.

So rather than play for a 15th season ? and his first outside the Steel City ? a tearful Ward opted to retire on Tuesday and secure a legacy unmatched in the franchise's long history.

"I can say I'm a Steeler for life and that's the bottom line, that's all I've really ever wanted," Ward said.

Ward holds every significant franchise receiving record, including receptions, receiving yards and touchdowns. His 1,000 career catches rank eighth all time and he is one of two players with at least 1,000 receptions and two Super Bowl rings.

The decision comes three weeks after the 36-year-old was released by the Steelers in a salary cap maneuver. Ward says he was contacted by several clubs but never had any formal discussions. He insists there are no hard feelings for his release, understanding that football is a business. As if to prove the point, Ward embraced Steelers owner Art Rooney II after stepping away from the podium following the announcement.

"Thank you (Mr. Rooney) for giving a small town boy from Forest Park, Ga., a chance," Ward said.

The former third-round pick out of Georgia was due to make $4 million next season, an expensive option for a player whose role diminished significantly in 2011 when he finished with 46 receptions, the fewest since his rookie season in 1998. He embraced his role as mentor to Pro Bowlers Mike Wallace and Antonio Brown even though he knew they were chewing into his playing time.

"I know the wideouts are going to be in great hands," he said. "They're full of talent."

And they're part of an offense that didn't exist when Ward made his debut 14 years ago. He spent most of his first three seasons blocking for running back Jerome Bettis, something he did better than any receiver in the league.

Over time, the Steelers evolved from the grind-it-out attack that has been the club's identity for decades. Ward's breakout season came in 2001 when he set a franchise record with 94 receptions then obliterated that mark in 2002 when he finished with 112 catches.

He made four straight Pro Bowls from 2001-2004 and seemed to get better as he aged. He was named the Most Valuable Player of the 2006 Super Bowl after catching five passes for 123 yards and a touchdown in Pittsburgh's 21-10 victory over Seattle, the franchise's first championship in 26 years. The Steelers added a second title in 2009 to give them six, more than any other team in the league.

Ward hoped to get the Steelers their seventh Lombardi Trophy but didn't catch a pass in a 29-23 overtime loss to Denver in the wild card round of last year's playoffs. Only one pass came his way, a dart down the sideline by quarterback Ben Roethlisberger during Pittsburgh's final drive in regulation. Denver cornerback Champ Bailey swatted the ball to the ground and Ward walked off the field and into the unknown.

The former "Dancing With the Stars" champion could have a lucrative postseason career in front of a camera ? he worked the red carpet during the Oscars ? but he maintained after his release he could still contribute. He still does.

"I feel like I have a few more good years in me left, Ward said. "I would love nothing more to get back to the Super Bowl."

He wasn't willing to do it, however, outside Pittsburgh.

"I want to go down as one of the greats to wear the black-and-gold and that's how it should end," Ward said.

Ward laughed when asked if he could go into coaching one day, taking a jab at coach Mike Tomlin, who isn't sure how Ward's passion would play in the locker room. One of the most respected players in the league because of his contributions on and off the field, Ward leaves a void that will be difficult to fill.

"On behalf of the NFL players, I want congratulate Hines on an extraordinary career," NFLPA executive director DeMaurice Smith said. "I know he will continue to be a leader and example to our men."

Ward's already started by urging Wallace to do what he can to remain with the Steelers.

"I told Mike you may get a chance to go other places but there's not another place like Pittsburgh," Ward said.

Women scored the last point in the ongoing contraception war with the failure of the Senate "Blunt amendment." It would have allowed any insurance agency or employer to deny women contraception if they find the coverage "morally objectionable."
?You'd think this would be the end of it since most Americans polled agree that throwing your own religious dogma into your customers' faces isn't right. Now some states are proposing similar legislation that say women would have to provide proof to providers and employers that the contraception they want is solely for medical purposes and is not to prevent pregnancy.
?This leaves women to do one of two things besides paying out of pocket on top of insurance premiums. For one, they can convert to whatever brand of Christianity their provider or employer is clearly suggesting they follow. In doing so, they accept that sex is not to be enjoyed or engaged in unless an 18+ year commitment comes out of it. Alternately, they can stick to paying for over-the counter methods, cross their fingers (or legs) and possibly spend much more (possibly government) money on an unwanted pregnancy.
?Do men have to have a "moral" reason to get Viagra from the same providers? Under current logic, erectile dysfunction should be treated with Viagra for a medical reason and if the resultanting erection and sex will lead to children. We must consider ED is God's way of saying, "You won't be needing that because I don't want you getting someone pregnant," and let guys deal with it.
?The Catholic church says new mandates would force health plans to "provide coverage of sterilization and contraception ? including abortifacient drugs."
Wrong. It doesn't cover anything but contraception. They also say contraception is inexpensive, so if we want to be heathens, do it on our own dime. Wrong again. I can't imagine any heathen ? I mean woman ? who may want to have sex more times than the number of children they want in a lifetime can afford the $30 to $90 uncovered visits a month on top of their insurance premiums to companies that refuse to cover contraception. Doesn't the Catholic church trust their flock to just say no?
I highly doubt that every man who is opposed to providing contraception through employers and insurance has only had sex to make his few children. They're all hypocrites.
Except Rush Limbaugh, who is simply stuck in a 6th grade mentality thinking if he calls grown women who are having sex "sluts? and "prostitutes" that it will simply shame them in to not having biological urges, or at least just to wait until called on by men who have them.
Rush, the amount of sex a woman has does not affect the cost of birth control. Republicans, do you really want Rush Limbaugh representing you? A man who says that taxpayers are pimps or johns if their wives, mothers, sisters, daughters and neices have more access to birth control and other important aspects of women's health?
The real issue is making contraception available to all women. Whoever chooses to use them does not follow the outdated and biologically improbable demands of their church, politician or Rush Limbaugh (whatever vile, sickening misogynist category he falls into). Well, that's on them. ?
